The insert contains BalI, AccI (3), BglII (2), SacI (2), EcoRI (2), and XbaI sites. TaqI RFLP best detected using a 1.1 kb BglII/HindIII segment of the insert, because the 3.8 kb HindIII insert contains a repetitive sequence. Enzyme(s) not detecting polymorphism (in a panel of 5): BamHI, BglII, EcoRI, HindIII, HincII, MspI, PstI, RsaI. Enzymes not detecting polymorphism: PstI, EcoRV, BamHI, BglII, MspI, EcoRI, HindIII, PvuII (10 unrelated Caucasians). Using this probe on TaqI digested human DNA, three RFLP alleles were detected.
