Restriction digests of the clone give the following sizes (kb): BamHI--5.4, 4.5; AvaI--9.9; HindIII--5.7, 3.4, 0.8; PstI--6.6, 3.3; EcoRI--5.4, 4.5. Contains internal BamHI (0.67 kb) and HindIII (1.2, 1.9, 5.2 kb) sites with distances relative to end closer to ampicillin site. The insert detects a 6.8 kb EcoRI genomic fragment. Recorded as not polymorphic by Collaborative Research, meaning either not polymorphic in 0.8% agarose gels, or difficult to work with. This probe did not detect previously reported SstI RFLPs when the genomic DNA was cut with SacI (an isoschizomer of SstI). This probe was derived from lambda ets H-3. Enzyme(s) not detecting polymorphism: BglI, HaeII, EcoRI, HindIII, MspI, HaeIII, HinfI, HincII, StyI, PstI, PvuII, BglII, BamHI. |
