Restriction digests of the clone give the following sizes (kb): EcoRI/HindIII--3.0 (doublet); BamHI--3.0, 2.0, 1.0; SalI--6.0; XhoI/PstI--3.0, 1.8, 0.55 (doublet); XbaI--3.0 (doublet). This was constructed from 3 overlapping cDNAs. When transcribed and translated in vitro, a polypeptide of 80 kDa recognized by anti-PFKL antibodies is generated. The clone detects the following genomic restriction fragments (kb), with those not from PFKL indicated by a '*': BamHI--8.6, 7.5, 5.0, 4.2*, 3.9*, 3.3, 2.9, 2.2*; KpnI--14, 11, 4.2, 3.3*, 2.6; HindIII-- >20, 12.3, 5.2. A fragment specific to PFKL can be generated from 3' sequences. The insert contains the complete coding sequences, with 5' and 3' untranslated sequences. The KpnI polymorphic site is located in the 3.3 kb BamHI genomic fragment . |
