Restriction digests of the clone give the following sizes (kb): Sal--uncut; BamHI/SalI--3.8; SalI/EcoRI--3.8; HindIII/EcoRI--3.0, 0.5, 0.3; AccI/AvaI--3.8; EcoRI/PstI--2.8, 0.9; EcoRI--3.8. Codominant segregation of the MspI RFLP was observed in 3 Caucasian families (9 individuals). The insert contains a portion of intron 1 which separates the exons coding for amino acids 1-21 and 22-79 of the gene. Codominant segregation of the TaqI RFLP was demonstrated in 5 families of 26 individuals. Enzyme(s) not detecting polymorphism: AluI, ApaI, BamHI, BglI, BglII, Bsp1286, DraI, EcoRI, HaeIII, HincII, HinfI, PstI, PvuII, ScaI, SphI, SstI, StuI, XbaI (tested against 7 unrelated North American Caucasians). |
