Restriction digests of the clone give the following sizes (kb): HindIII/XbaI--4.2, 3.1; BamHI--3.4, 2.0, 1.5, 0.41; PstI--7.2; SacI--7.2; PvuII--2.7, 2.45, 1.10, 0.56, 0.28. Constructed from a 520 bp HindIII/PstI PCR product and the large PstI/EcoRI fragment from HK-3.5. The SmaI site was lost during cloning, and the vector SacI site was deleted. The insert contains the following restriction sites (approximate kb from the 5' end): BamHI--0.38, 2.3, 3.7; PstI--0.52; SacI--1.1; PvuII--2.1, 2.6, 2.9, 3.9. |
